Working Principle of UV Visible Spectrophotometer

Different matter has different but special absorbance wavelength point. Also, when at the fixed wavelength point, the absorbance has some relation to the substance’ s (Always transparent Solution) concentration and its thickness. The relation can be concluded as the following Formula which is called Lambert-Beer Law.

  • T= I/Io
  • A=KCL= -log I/Io
  • A   Absorbance
  • C   Concentration of the Solution
  • K   Absorbance Coefficient of the Solution
  • L   The length of the Solution in the light path
  • I   The intensity of the light focused on the A/D after it permeate the solution to be measured.
  • Io   The intensity of the light focused on the A/D after it permeate the Solution.

Note: When test, the solvent is usually taken as the Reference Solution and its Transmittance is considered as 100%T. While the Transmittance of the sample to be tested is a relative value which is got comparing to that of the Reference.

FAQ

No. The DV-8200 is a Visible Spectrophotometer with a wavelength range of 320–1100 nm. It is designed for colorimetric analysis of visible-light absorbing samples only and is not suitable for measurements requiring UV (<320 nm) detection.

Yes. The instrument features a built-in Standard Curve function. After zeroing with a blank and measuring known standard concentrations, the DV-8200 constructs a calibration curve and directly displays the concentration of unknown samples.

No. The DV-8200 can be operated standalone using the built-in keypad and large LCD display for wavelength selection, blanking, and direct reading. PC connectivity (optional) is only needed for advanced functions such as full wavelength scanning, kinetics, and enhanced data processing.

The DV-8200 accepts standard 10 mm pathlength glass or quartz cuvettes (glass cuvettes are recommended for the visible range). It is also compatible with various rectangular cuvette holders for different sample volumes.

Yes. The DV-8200 is equipped with RS-232 and USB ports, allowing direct connection to a serial printer for hard copy results, or data export to a PC via USB (requires optional PC software).

The DV-8200 is ideal for teaching laboratories in colleges and universities, environmental water testing stations, food & agricultural QC labs, pharmaceutical QC departments, and small-to-medium industrial quality control labs requiring routine visible-range colorimetric analysis.

The optional PC software, connected via USB, unlocks advanced features including full wavelength scanning, kinetics testing, DNA/protein concentration measurement (A260/A280), and enhanced standard curve fitting. These capabilities significantly expand the flexibility and efficiency of data analysis.
