The most common issues are contamination of the guard column or analytical column frit, clogging of the in-line filter, or partial blockage of the pump head inlet filter due to particulates in the mobile phase.

Disassemble the individual units (such as the chromatographic column, guard column, and in-line filter) one by one to identify the specific component causing the blockage. For in-line filters, the filter element can be replaced directly; for check valves and filter heads, ultrasonic cleaning with ultrapure water and dilute nitric acid can be used.

First, ensure the eluent is thoroughly degassed. If the issue originates in the detector, flush it by injecting ultrapure water and methanol via a syringe to expel air bubbles. For regular fluctuations caused by the high-pressure pump, inspect and clean the check valves.

Peak tailing usually indicates column contamination or a spent guard column. Peak broadening may result from excessive system dead volume—for instance, due to overly long connecting tubing or mismatched internal diameters.

Recommendation: First, try replacing the guard column. If the problem persists, the column requires cleaning and regeneration. Additionally, inspect the tubing between the injection valve and the detector; PEEK tubing with the smallest possible internal diameter (e.g., 0.25 mm) should be used to minimize dead volume.

Use freshly prepared eluent and install a CO₂ trap on the eluent bottle to prevent CO₂ absorption. Additionally, ensure consistency in the quality of the ultrapure water and reagents used for each preparation.

When a chromatography column is not in use for a short period, it should be flushed according to the manufacturer’s instructions and then sealed and stored in the recommended solvent.For long-term storage, the column ends must be tightly capped to prevent the mobile phase from evaporating and causing the column bed to dry out.

Inverted peaks typically occur due to contamination by ions—identical to those in the sample—present in the eluent or the flow path system. Thoroughly clean the eluent bottle and prepare fresh eluent using higher-purity reagents and ultrapure water.

Use a guard column: Installing a guard column before the analytical column and replacing it regularly is the most effective method.

Rigorous sample pretreatment: All samples must be filtered through a 0.45 μm membrane or processed using online ultrafiltration/dialysis techniques prior to injection to remove particulates and impurities.

Proper usage: Avoid using eluents that fall outside the column’s pH range (typically 2–11) and ensure the use of high-purity reagents.

The system supports 2, 3, 4 and 5 mm i.d. columns, with up to a 250 mm analytical column plus 50 mm guard column.

The built-in 3D constant-temperature system has a stated stabilization time of less than 30 minutes.
